Four runners were together in the closing stages of the race that involved a 1.5-kilometre swim, 40km cycle and 10km run but Frodeno powered to victory in the final 300 metres to win in a time of 1 hour 48 minutes 53 seconds.
Simon Whitfield of Canada, the Olympic champion from Sydney 2000, took silver with bronze going to New Zealand's Bevan Docherty, a silver medallist in the event four years ago.
World champion and race favourite Javier Gomez of Spain trailed in fourth.
